溴氰菊酯对德国小蠊碱性磷酸酶抑制效应

Inhibitory effect of deltamethrin on alkaline phosphatase in Blattella germanica

      目的  通过研究溴氰菊酯与德国小蠊碱性磷酸酶(AKP)活性水平的量效与时效关系,进一步完善溴氰菊酯作用机制,为德国小蠊的防制工作提供理论基础和参考方法。
      方法  参照国标GB/T 13917.1 – 2009《农药登记用卫生杀虫剂室内药效试验及评价》喷施一定剂量不同浓度的溴氰菊酯,用AKP测定试剂盒测定德国小蠊AKP活性。
      结果  72 h内AKP的活性水平从对照组到5组随溴氰菊酯浓度的增大表现为显著持续抑制作用。施药72 h,2、4、5组抑制率分别达到本组最高点23.9 %、29.5 %和27.3 %;施药96 h,1和3组抑制率分别达到本组最高点19.9 %和26.8 %。
      结论  德国小蠊AKP是溴氰菊酯的作用靶标之一,由于AKP活性敏感度降低而产生的靶标抗性是德国小蠊对溴氰菊酯产生抗药性的原因之一,德国小蠊AKP活性水平可作为对拟除虫菊酯类杀虫剂抗药性水平监测的指标之一。

     

    Abstract:
      Objective  To explore dose- and time-effect of deltamethrin (DM) on alkaline phosphatase (AKP) activity in Blattella germanica and to provide references for the control of Blattella germanica.
      Methods  According to the national standard described in Efficacy Test and Evaluation Methods for Indoor Application of Hygienic Insecticide under Pesticide Registration (GB/T 13917.1 – 2009), totally 300 healthy post-emergence Blattella germanica of sensitive strain were randomly divided into 5 treatment groups exposed to different sublethal concentrations (1.54, 3.07, 6.14, 12.28, and 24.56 mg/L) of anhydrous acetone DM solution spray and a control group with only anhydrous acetone (50 in each group). The activities of AKP in Blattella germanica at 24, 48, 72, 96, 120, and 144 hours of the treatment were determined by using AKP Kit.
      Results  Dose-dependently and steadily inhibitive effect of DM on KAP were observed in Blattella germanica of all the 5 treatment groups with 72 hours. The maximum inhibitive rates were 23.9%, 29.5%, and 27.3% under the dosages of 3.07, 12.28, and 24.56 mg/L at 72-hour and the rates were 19.9% and 26.8% under the dosages of 1.54 and 6.14 mg/L at 96-hour after the DM treatment.
      Conclusion  The activity of AKP in Blattella germanica could be used as one of the indices for determining resistant level of Blattella germanica to pyrethroid insecticides because of the resistance of Blattella germanica to deltamethrin is due to the decrease in AKP sensitivity to the insecticide.
