镧对仔鼠大脑海马星形胶质细胞兴奋性影响

Effect of lanthanum on excitability of hippocampus astrocytes in offspring rats

      目的  观察氯化镧(LaCl3)对星形胶质细胞兴奋性的影响,探讨其神经毒作用机制。
      方法  雌性Wistar大鼠40只,随机分为5组,对照组饮用蒸馏水,染镧组分别饮用含有不同浓度氯化镧(0.125 %、0.250 %、0.500 %和1.000 %)的蒸馏水溶液,从雌鼠妊娠起到仔鼠断乳后1个月结束。采用Fura-2荧光探针法检测仔鼠大脑海马神经细胞内Ca2+浓度;酶联免疫分析(ELISA)法测定1,4,5 – 三磷酸肌醇(IP3)含量;实时定量PCR法测定促代谢性谷氨酸受体5(mGluR5)、磷脂酶C(PLC)和缝隙连接蛋白(Cx43)mRNA转录水平;Western blot法测定mGluR5、PLC和Cx43蛋白表达水平。
      结果  与对照组比较,随氯化镧染毒剂量增加,染镧组仔鼠海马神经细胞内Ca2+浓度与IP3含量趋势性升高,呈剂量效应关系(P < 0.05);与对照组比较,各剂量染镧组大鼠仔鼠海马神经元内mGluR5、PLC和Cx43 mRNA表达上调,mGluR5、PLC和Cx43蛋白表达水平显著上调,呈剂量效应关系(P < 0.05)。
      结论  氯化镧可致大鼠仔鼠脑海马星形胶质细胞过度兴奋,诱发兴奋性毒性,进而造成神经元损伤。

     

    Abstract:
      Objective  To observe the effect of lanthanum chloride (LaCl3) on excitability of hippocampus astrocytes and to explore the mechanism of the neurotoxic effect.
      Methods  Forty female Wistar rats were randomly divided into 5 groups; the rats of the 4 experimental groups were supplied with distilled drinking water containing LaCl3 at various concentration (0.125%, 0.250%, 0.500%, and 1.000%) and those of the control group with uncontaminated distilled water during the period from pregnancy till the end of first weaning month. Intracellular bivalent calcium ion (Ca2+) concentration in hippocampal neurons of the offspring rats was detected with Fura-2 fluorescent probe method and the inositol 1,4,5-trisphosphate (IP3) content was determined with enzyme-linked immunosorbent assay (ELISA). The mRNA transcription of metabotropic glutamate receptor 5 (mGluR5), phospholipase C (PLC), and connexin (Cx43) were determined with real-time quantitative PCR; the protein expression of mGluR5, PLC, and Cx43 were detected with Western blot.
      Results  Compared with those of the control rats, the Ca2+ concentration and IP3 content in hippocampal neurons of the experimental rats increased with the increment of LaCl3 in a dose-effect manner (both P < 0.05); while, the expressions of mGluR5, PLC, and Cx43 mRNA and protein in hippocampal neurons of experimental rats were all up-regulated with the increment of LaCl3 in a dose-effect manner (P < 0.05 for all).
      Conclusion  Lanthanum chloride can cause excessive excitability of hippocampal astrocytes and induce neuronal damage in offspring rats with maternal Lanthanum chloride exposure.
