非综合征型唇腭裂患者父母ROR2基因多态性分析

MAFs and haplotype frequencies for SNPs in ROR2 gene among parents of Han Chinese NSCL/P patients in China

  • 摘要: 目的分析和比较中国山东、武汉2地区汉族非综合征型唇/腭裂(NSCL/P)病例父母受体酪氨酸激酶样孤立受体2(ROR2)基因单核苷酸多态性(SNPs)的最小等位基因频率(MAF)及单体域。方法采用Haploview软件对Hardy-Weinberg平衡且各地区间最小等位基因相同的非纯合SNPs进行MAF及频数分布的分析,用MAF差值相对比指标比较地区间MAF水平的异同;并对2地区基因型测定率>80%的非纯合SNPs进行单体域及单体型频率分析及比较。结果2地区所测 101个SNPs中,89个非纯合SNPs的最小等位基因相同,MAF差值相对比<20%的占76.4%,≥50%的占4.5%,且均位于第1内含子区;与山东地区比较,基因型测定率>80%的94个非纯合SNPs在武汉地区形成了1个位于第1内含子区的独立单体域。结论山东和武汉地区研究人群所测SNPs的特征以共性为主,MAF差值相对比≥50%的SNPs和武汉地区的独立单体域均位于第1内含子区。

     

    Abstract: ObjectiveTo analyze minor allele frequencies(MAFs) and haplotype frequencies for single neucleotide polymorphisms(SNPs) in receptor tyrosine kinase-like orphan receptor 2(ROR2) gene among parents of patients with non-syndromic cleft lip with or without cleft palate(NSCL/P) trios recruited from Shandong province and Wuhan city in China.MethodsMAF and its frequency distribution were analyzed for SNPs in ROR2 gene according to Hardy-Weinberg equilibrium of recruitment site with Haploview program.The difference in relative ratio of MAF was used to identify the MAF difference between two sites.Haplotype blocks and frequencies were analyzed and compared for SNPs of MAF>0.01 and genotyping rate>80%.ResultsAmong 89 non-homogenous SNPs with same minor allele in 101 SNPs genotyped,the proportions for those with different relative ratio less than 20% and equal or greater than 50% were 76.4% and 4.5%,respectively.The four SNPs with different relative ratio of MAF ≥50% were located in the first intron region.Compared with that of Shandong,the 94 non-homogenous SNPs with genotyping call rate >80% formed an independent haplotype block in the first intron region in Wuhan.ConclusionThe SNPs for ROR2 genotyped in Shandong and Wuhan present similar distribution,with the four SNPs with different relative ratios equal or greater than 50% in the two areas and one independent haplotype block locating in the first intron region identified in Wuhan.
