原发性高血压与2种相关基因多态性关系

Relationship between essential hypertension and two related gene polymorphism

      目的  探讨18~45岁人群的原发性高血压(EH)与G蛋白β3亚单位(GNB3)基因825C/T、Y染色体HindIII酶切位点多态性的关系。
      方法  选取原发性高血压患者345例, 正常人281名。提取白细胞DNA, 采用多聚酶链反应结合限制性内切酶(BseDI或HindⅢ)方法检测2种基因的多态性特征。
      结果  对照组和病例组间GNB3825C/T多态性各基因型和等位基因频率差异无统计学意义(P > 0.05)。Y染色体HindIII多态性各基因型差异有统计学意义(P=0.03), EH病人HindIII(-)基因型增多。
      结论  Y染色体HindIII酶切位点多态性与青年原发性高血压相关。

     

    Abstract:
      Objective  To investigate the relationship of GNB3 825C/Tpoly mor phism and polymorphic HindIIIrestriction site of the Ychromosome in young aged essential hypertension.
      Methods  345 patients were enrolled with essential hyper tension and 281 healthy subjects as control group.Genomic DNAwas extracted from white blood cell.Segments of GNB3 and Ychromosome were amplified from genomic DNAby polymerase chain reaction(PCR).PCRproducts were subjected to electrophoresis in agarose gels.PCRproducts were restricted with HindIIIor BseDIrespectively.The digested products were subjected to electrophoresis in agarose gels and stained with ethidium bromide.
      Results  GNB3 825C/Tallele and genot ype fr equencies did not differ significantly between essential hyper tension and control group(P>0.05).The HindIII(+)genotype was found in 55.4% of the men in hyper tension and in 67.3% of those in contr ol.The frequency of HindIII(-)genoty pe was sig nificantly higher than that in the contr ol(P=0.03).
      Conclusion  Polymorphic HindIIIr estr ict ion site of the Ychromosome maybe is associated with young essential hypertension.
