Effect of alcohol on cytokines in hepatic tissue

      目的  通过建立酒精性脂肪肝大鼠模型, 并给予辛伐他汀及非诺贝特对酒精性脂肪肝进行干预, 探讨转化生长因子β-1(TGF-β1)、血管内皮生长因子(VEGF)、血小板衍生性生长因子受体-β(PDGFR-β)在这一过程中的变化, 以探讨酒精性脂肪肝的发病机制。
      方法  肝脏病理用苏木精-伊红(HE)染色, 光镜下观察。用免疫组化方法检测各种模型鼠肝中TGF-β1、VEGF、PDGFR-β的表达。
      结果  非诺贝特组VEGF表达无明显变化, 辛伐他汀组可见表达增加(P < 0.05);非诺贝特组肝组织TGF-β1及PDGFR-β表达减少; 辛伐他汀组则仅FDGFR-β的表达显著降低。
      结论  酒精干预后, VEGF、TGF-β1及PDGFR-β在肝组织中均呈阳性表达; 非诺贝特和辛伐他汀干预后可见其不同程度的减少, 且与病理的改善相平行。

     

    Abstract:
      Objective   To investigate the effect of the cytokine-vascular endothelial gro wth factor(VEGF), transform growth factor β1(TGF-β1), platelet-derived growth factor receptor-β(PDGFR-β)in the etiology of alcoholic fatty liver by setting up rat models of fatty liver, and also the changes of the cytokines in the progress of the treatment by simvastation and fenofibrate.
      Methods   Liver histological pathology was dyed by HE and analyzed under light microscope; liver immunohistochemical staining was performed by SABC technique to assess the cytokine-VEGF, TGF-β1, PDGFR-β in hepatic tissue.
      Results   It was found post-treatment with drugs that there was significant improvement in pathology of alcoholic fatty liver model after treated with fenofibrate, but not with simavastatin.There was no significant differences after treated with finofibrate(P > 0.05), but an improved level post treated with simav astatin(P > 0.05); after treated with fenofibrate, there was decreased expression of TGF-β1 and PDGFR-β, and a significant decrease of PDGFR-β after treated with simavastatin.
      Conclusion   Post-treated with alcohol, VEGF, TGF-β1 and PDGFR-β were all possitive expressed in alcoholic fatty liver tissue, and a decreased expression post-treated with fenofibrate and simavastatin, which was parallel with the changes of pathology, and illustrated cytokines played roles in the etiology of alcoholic fatty liver.
