糖尿病大鼠一氧化氮的变化及锌硒的保护作用

Changes of serum nitric oxide and protection of zinc and selenium in diabetic rats

  • 摘要:
      目的   研究一氧化氮(NO)在胰岛细胞病理损伤中所起的作用及锌、硒对胰岛氧化损伤的保护机制。
      方法   雄性Wistar大鼠42只, 随机留取12只为正常对照组, 其余大鼠以四氧嘧啶诱导成糖尿病模型, 随机分为糖尿病对照组和2个实验组(糖尿病补锌组、糖尿病补硒组)。每日灌胃补充一定剂量的锌、硒。实验期为60d。分别进行实验前后生化指标的检测。
      结果   糖尿病补锌组(DM+Zn)、补硒组(DM+Se)大鼠血糖水平较实验前有一定程度的降低; 血清胰岛素水平及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPX)活力有明显提高; 血清一氧化氮、一氧化氮合酶水平下降。
      结论   锌、硒通过提高抗氧化酶的活性、抑制一氧化氮的生成等途径减轻自由基对胰岛β细胞的氧化损伤。

     

    Abstract:
      Objective   To study the effects of zinc and selenium on serum nitric oxide and protection on oxidative damage of pancreatic islets in diabetic rats.
      Methods   12 rats were randomly selected from 42 male Wistar rats as normal control group (NC group).The rest induced by allox an into diabetic model were randomly divided into diabetic control group (DM group) treated with water and 2 other experimental groups (DM+Zn group, DM+Se group) supplemented with zinc, selenium respectively by gavage daily for 60 days. At the end of the treatment, the blood biochemical analysis were performed.
      Results   The blood glucose in DM+Zn group, DM+Se group decreased significantly than that before experiment, the levels of serum insulin and the activity of superoxide dismutase (SOD), glutathione peroxidase (GPX) were also improved obviously. Compared with DM group, the levels of serum nitric oxide (NO), nitric oxide synthase (NOS) in DM+Zn group and DM+Se group were significant lower.
      Conclusion   Zinc and selenium could alleviate oxidative damage on β cells of pancreatic islets caused by free radicals through improving the antioxidative enzyme activities and inhibiting the levels of serum nitric oxide in diabetic rats.
