碳酸缓冲液对大鼠光气肺损伤影响的研究

Study on therapeutical effect of carbonic acid buffer on rats exposed to phosgene and its mechanisms of oxidation

      目的   探讨光气肺水肿形成机理及碳酸缓冲液对大鼠光气中毒的影响。
      方法   40只SD大鼠被随机分为阴性对照组、阳性对照组、0.2ml碳酸缓冲液治疗组、0.5ml碳酸缓冲液治疗组.光气染毒后5min, 阳性对照组注射0.5ml生理盐水, 两个治疗组分别注射碳酸盐缓冲液(碳酸缓冲液: Na2CO3/NaHCO3pH: 76, 01mmol/L)0.2和0.5ml, 3h后处死采血并开胸取肺, 用荧光分光法检测脂质过氧化产物丙二醛(MDA)、还原型谷胱甘肽(GSH)和氧化型谷胱甘肽(GSSG), 测量并计算肺湿干比。
      结果   与阴性对照组相比, 光气染毒大鼠血清MDA含量显著升高(P < 0.01), 血清GSH显著降低(P < 0.01), 肝脏GSSG显著升高(P < 0.05), 肺湿干比增大(P < 0.05).与阳性对照组相比, 给予碳酸缓冲液治疗组血清MDA均显著降低, 05ml碳酸缓冲液对血清MDA的抑制作用更强(P < 0.05);两个碳酸缓冲液治疗组血清GSH升高显著(P < 0.05);治疗组的肺湿干比有所降低, 但无显著性差异(P < 0.05)。
      结论   光气中毒后出现氧化损伤, 抗氧化剂GSH损耗严重; 给予碳酸缓冲液可以缓解光气中毒肺损伤。

     

    Abstract:
      Objective   To study the mechanism of pulmonary edema formation and the effect of treatment with carbonic acid buffer on rats exposed to phosgene.
      Methods   40 SD rats were randomly divided into four groups: control group, poisoning group, 0.2 ml carbonic acid buffer group and 0.5 ml carbonic acid buffer group.At the fifth minute the treated groups(0.2 ml group and 05 ml group)were injected with 0.2 or 0.5 ml of carbonic acid buffer by abdominal cavity, respectively.At the third hour, all the rats were killed by decollation.The fluorescence-metry was employed to measure the MDA and GSH.Wet-to-dry weight ratio of rat lung was calculated.
      Results   Compared with the control group, the blood serum MDA level increased(P < 0.01)and GSH content decreased(P < 0.01), the GSSG of liver and the wet-to-dry weight ratio of lung lung increased(P < 0.05)in the poisoning groups.Compared with the poisoning group, the MDA levgel lowed(P < 0.01)and GSH content raised(P < 0.05)in the treated groups, the both changes of the MDA level and GSH content in 0.5 ml carbonic acid buffer group were more significant than those in the 0.2 ml carbonic acid buffer group.The wet-to-dry weight ratio of lung decreased in treated groups, but no significant difference was observed.
      Conclusion   The oxidative stress injury was one of mechanisms of phosegene toxicity in acute poisoned rats.The carbonic acid buffer may have the relieving role on the lung damage in rats exposed to phosgene.
