改良免疫-PCR检测肾综合征出血热特异性抗体

Study on modified immuno-PCR method for detection of HFRS antibodies

  • 摘要:
      目的   应用改良免疫PCR方法检测肾合征出血热(HFRS)特异性抗体。
      方法   在紫外线照射处理后的PCR管中进行免疫PCR反应, 以链霉亲和素作为连接分子连接生物素标记二抗和生物素标记DNA, PCR扩增DNA分子, 琼脂糖电泳及扫描判断结果。
      结果   改良免疫PCR方法的阳性检出率为78.5%, 常规免疫PCR法和ELISA法的阳性检出率分别为62.3%和47.4%。
      结论   改良免疫PCR方法对出血热特异性抗体的阳性检出率高于常规的免疫PCR法和间接ELISA法, 可用于肾综合出血热特异性抗体的检测。

     

    Abstract:
      Objective   To detect HFRS antibodies by modified immuno-PCR method.
      Methods   Immuno-PCR reaction was operated in PCR tubes, which were treated with the shining of UVray, and streptavidin was used to bridge the biotinylated antibodies and biotinylated DNA, then the DN A was amplified in the same PCR tube.The PCR production was analyzed in agarosegel.
      Results   The positive rate of specific antibodies by modified immuno-PCR was 78.5%, that detected by routine immuno-PCR and indirect EL ISA method were 62.3% and 47.7% respectively.
      Conclusion   The positive rate of specific antibodies by modified immuno-PCR was higher than that of routine immuno-PCR and indirect EL ISA methods, so modified immuno-PCR might be used to detect HFRS antibodies.
