黄芪多糖对大鼠β-防御素基因表达的调节作用

Regulation of rBD-2 gene expression by asttagalus polysaccharides

      目的   探讨黄芪多糖对大鼠呼吸系统防御素表达的调节作用。
      方法   将60只健康Wistar大鼠随机分成对照组和实验组。实验组经黄芪多糖溶液灌胃; 对照组以等量生理盐水(NS)灌胃, 分别于第0h, 第1, 3, 5, 7d处死, 提取其肺组织总RNA, 用RT-PCR法检测大鼠β-防御素-2基因的表达变化。
      结果   黄芪多糖实验组β-防御素-2mRNA的表达高于对照组(P < 0.05), 有显著性差异; 且实验组中, 第3dβ-防御素-2mRNA表达水平高于第1, 5, 7d时的水平(P < 0.05), 有显著性差异。
      结论   黄芪多糖可以上调大鼠肺组织rBD-2mRNA的表达。

     

    Abstract:
      Objective   To explore the regulation of rBD-2 gene expression by Astragalus Polysaccharides (APS).
      Methods   Sixty Wistar rats weighing 150 g were randomly divided into the control group and the APS group with 30 rats each. In APS group, each was dealt with APS ig; A similar dose of NS was given to the rats of the control group. Lungs were harvested on 0h, and in 1, 3, 5 and 7 d respectively after APS stimulation. Total RNA was extracted from tracheal and pulmonary tissue and rBD-2 was measured by R T-PCR.β-actin was used as internal standard.
      Results   The rBD-2 mRNA expressio in APS group, showed a marked increase as compared with that in the control group, and also on 3 d, compared with 1, 5 and 7 d after APS stimulation.
      Conclusions   Astraglus polysaccharides could up-regulate rBD-2 mRNA in rat lung tissue.
