铅对发育期小鼠脑一氧化氮合酶1表达的影响

Effect of lead on NOS1 expression in brain of developmental mouse

  • 摘要:
      目的   探讨不同浓度铅对发育期小鼠脑中一氧化氮合酶(NOS)表达的影响及其与小鼠学习记忆功能的关系。
      方法   分别于P1生后1d, P8, P15, P22, P30等5个时间段取不同浓度染铅的幼鼠脑海马组织进行蛋白免疫印迹实验。
      结果   对照组小鼠脑中一氧化氮合酶1从P1至P30逐渐降低, 低浓度染铅(0.5mmol/L)组个别时段NOS1升高, 较高浓度染铅(2.4~7.2mmol/L)组有较多时段升高, 但更高浓度的染铅(9.6~14.4mmol/L)组NOS1表达逐渐降低。
      结论   染铅小鼠脑NOS1表达与未染铅组比较有明显变化, 表明NOS1可能与小鼠学习记忆功能发育有重要关系。

     

    Abstract:
      Objective   To study lead effect of different concentrations on the expression of Nitric Oxide Synthase(NOS1) in mouse brain of developmental period and relation with learning memory function of mouse.
      Methods   To take out the brain tissue of mouse that exposed to differ ent concentration of lead in 5 time segments, such as, P1(postnatal 1d), P8, P15, P22, P30 etc.respectively dividid into groups after homogenate and then proceed to Westem blot experiment.
      Results   The expression of NOS1 in mouse brains of control group decreased gradually from P1 to P30;and in the group of low concentration of lead exposure(0.5 mmol/L), NOS1 increased in a few time segment; and in the groups of higher concentration of lead exposure(2.4-7.2 mmol/L), the expression of NOS1 increased in more time segments; but in the highest concentration groups (9.6-14.4 mmol/L), the NOS1 expression decreased gradually.
      Conclusion   The expression of NOS1 in lead-exposed young mouse brain were different to that in the groups without lead exposure, which indicated that NOS1 may have important relation with development of learning memory function.
