赤潮藻细胞计数方法比较研究

Comparative study on cell count method with prorocentrum donghaiense

摘要:
目的建立简便、快捷、准确、可行的赤潮藻细胞计数方法。
方法采用显微镜法、可见分光光度法、库尔特法、流式细胞计数显微镜(Flow CAM)等方法对纯培养的东海原甲藻进行细胞计数。
结果 3种细胞计数法与显微镜计数相比较, 均有较好的线性相关性。
结论 4种细胞计数方法各有其优缺点, 其中库尔特计数是相对简便、快捷、准确、可行的细胞计数方法。

Abstract:
Objective To found simple, exact and feasible cell count methods.
Methods Microscope, visible spectrophotography, Flow CAM(Flow Cytometer and Microscope)and Multisizer^TM3 Coulter counter were used to measure cell amount of prorocentrum donghaiense.
Results The insightful comparison on the interrelations of the above 3 mentioned cell count methods was made with the method of microscope, which revealed the sound linear interrelations between them.
Conclusion All the 4 mentioned cell count methods had its advantages and disadvantages, among which, the cell count method with Multisizer^TM3 Coulter counter was concise, exact and reliable experimental approach.
