螨媒传播汉坦病毒的实验研究

Experimental study on role of mites in transmission of Hantaan virus

  • 摘要:
      目的   研究革螨、恙螨在肾综合征出血热(hemorrhagic fever with renal syndrome, HPRS)传播中的媒介作用。
      方法   采集自然界鼠窝革螨及饲养繁殖的子代革螨和鼠体恙螨幼虫以及从小黑板采集游离恙螨幼虫及饲养孵化的若虫; 进行螨细胞培养分别观察螨细胞传播汉坦病毒(Hataan virus, HV)的效能。
      结果   发现螨细胞培养第15d和第20d可从螨细胞内检测到HV; 其中HV检出数和荧光强度在革螨子4代、子3代和恙螨若虫均多于和强于子1代、子2代及恙螨幼虫。经原位RT-PCR检测, HV-RNA在革螨子3代、子4代和恙螨若虫细胞内的阳性颗粒较革螨子1代、2代和恙螨幼虫多且密集。
      结论   革螨、恙螨对HV的传播有一定的媒介作用, 在维持HV的自然循环中起重要作用。

     

    Abstract:
      Objective   To study the roles of gasmid mite and chigger mite in the transmission of Hantaan virus(HV).
      Methods   Natural gasmid mite, the first generation gasmid mite, chigger mite larvas from rats, free chigger mite larva and nymph were collected and their cells were cultured to study the role of transmission HV.
      Results   HV was detected from mite cells after 15 and 20 days of cultured.The number of HV and fluorescence intensity detected from the third and fourth generation gasmid mite and chigger mite nymph were much more than those from the tfirst, second generation and chigger mite larva.The signal particles in the third and fourth generation cells of gamasid mites and nymph cells of chigger mites were more than that in the first and second generation cells of gamasid mites and larva cells of chigger mites.
      Conclusion   Gasmid mite and chigger mite had an important role in playing on HV transmission in keeping natural cycles.
