洛伐他汀对HL-60细胞bcl-2基因表达的影响

Effects of lovastatin on expression of bcl-2 in HL-60 cells

      目的   观察HMG-CoA还原酶抑制剂洛伐他汀(lovastatin, LOV)对HL-60细胞bcl-2基因表达的影响, 进一步探讨其抑制细胞增殖诱导细胞凋亡的可能分子机制。
      方法   采用生长曲线测定、流式细胞仪分析、DNA片段化百分率检测以及RT-PCR、Westernblot等技术方法, 观察LOV对HL-60细胞体外增殖和凋亡的影响, 以及bcl-2基因表达的改变。
      结果   LOV能显著抑制HL-60细胞增殖, 使细胞周期阻滞于G0/G1期, 细胞凋亡率增加, DNA片段化百分率升高; LOV能下调HL-60细胞bcl-2mRNA及蛋白的表达。
      结论   LOV对HL-60细胞有增殖抑制和诱导凋亡作用, 下调节bcl-2基因表达可能是其分子机制之一。

     

    Abstract:
      Objective   To study the effects of lovastatin(LOV)on the expression of bcl-2 and proliferation and apoptosis of HL-60 cells, and its possible molecular mechanisms.
      Methods   The cellular proliferation capacity was observed with the growth curve assay, DNA fragmentation rate was used to detect apoptotic changes.The cell cycle distribution and apoptotic rate were measured with flow cytometry.the expression of bcl-2 mRNA was analyzed with RT-PCR.The protein expression of bcl-2 was detected by Western blotting.
      Results   The proliferation capacity was decreased in cells treated by LOV, the apoptotic rate, the DNA fragmentation rate were increased and the expression of bcl-2 was decreased in cells treated by LOV.
      Conclusion   Our study demonstrated that LOV can inhibit cell growth and induce apoptosis.The down-regu2 lated expression of bcl-2 maybe play a key role in the synergistic anti-tumor of LOV.
