河蚬糖蛋白对人肝癌细胞凋亡的影响

Effect of glycoprotein from corbicula fluminea on apoptosis of BEL7404

  • 摘要:
      目的   探讨河蚬中提取的糖蛋白(CFp-a)的体外抗癌作用。
      方法   体外培养人肝癌细胞BEL7404, 利用透射电镜、体外细胞毒性试验(MTT)法、流式细胞仪观察CFp-a在体外对BEL7404细胞增殖和诱导细胞凋亡的作用。
      结果   CFp-a在体外具有抑制BEL7404细胞增殖的作用。透射电镜观察到CFp-a处理细胞产生凋亡形态学改变。在DNA直方图上观察到典型的亚“G1”峰。CFp-a对细胞周期中的G1-S和G2-M期两个检查点均有影响。
      结论   从河蚬中提取的糖蛋白CFp-a对BEL7404细胞具有明显诱导BEL7404细胞凋亡作用。

     

    Abstract:
      Objective   To study the antitumor effect of the glycoprotein CFp-a from corbicula fluminea against BEL7404 cancer cells in vitro.
      Methods   The inhibition rates of cancer cell was measured by MTT method.The morphological change of BEL7404 cell was detected by electronic microscopy.The effect on cell cycles of BEL7404 were observed by FCM.
      Results   The glycoprotein CFp-a had strong antitumor effect and induced apoptosis in vitro.The morphological change of apoptosis cell was also founded by electronic microscopy.A typical subdiploid peak sub G1 appeared on DNA histogram in FCM.CFp-a affected both G1-S and G2-M checkpoints.
      Conclusion   The glycoprotein CFp-a from corbicula fluminea can induce apoptosis of BEL7404 cell.
