刺五加对放射损伤骨髓微血管的保护作用

Protection effects of injection acanthopanax senticosus on bone marrow hematopoietic microenvironment in mice with the radiation damage

  • 摘要:
      目的   观察刺五加注射液对放射损伤小鼠骨髓微血管的保护作用。
      方法   用60Coγ射线辐射小鼠建立骨髓微血管放射损伤模型; 6周龄雌性小鼠随机分为正常、模型、模型+刺五加注射液低剂量, 模型+刺五加注射液高剂量和模型+复方丹参注射液5组。采用分光光度法分别测定各组小鼠股骨骨髓血红蛋白溶液标本的吸光度(A)值; 鼠尾采血测定外周血红细胞、白细胞数量和血红蛋白含量; 并分别观测骨髓毛细动脉根数、面积和骨髓造血组织容量百分率。
      结果   刺五加注射液能明显降低放射损伤小鼠骨髓血红蛋白含量, 提高外周血红细胞和白细胞的数量以及增加骨髓毛细动脉根数、面积和骨髓造血组织容量百分率。
      结论   刺五加注射液能明显修复放射损伤小鼠骨髓微血管, 保护骨髓造血微环境。

     

    Abstract:
      Objective   To observe the protective effect of injection of acanthopanax senticosus on the bone marrow hemato poietic microenv ironment in mice with the radiation damage.
      Methods   (1) The model of the mice.damaged bone marr ow micrang ium was made by whole-body irradiation with 60Coγ-radiation.(2)Mice(female)aged 6 weeks were randomly divide into the normal group, the model group, the model plus the inject ion of acanthopanax senticosus(high and low doses; 0.2 mg/10g, 0.4 ml/10g)as well as positive control group(model plus injection salvia miltiorrhiza).Mice's thigh bone marrow hemoglobin solution specimen were taken respectively to test optical density(OD)by spectrophotometry.The peripheral blood specimen were taken respectiv ely by mice-tail to test RBC, WBC and Hb quantity.To observe respectively the number of the bone marrow arteriocapilly, its area and the percentage of the capacity of hemapoictic tissue.
      Results   The injection of acanthopanax senticosus can remar kably decrease the bone marrow hemoglobin content and the number of the peripher al blood RBC, WBC and increase the number of bone marrowar teriocapillary, its area and raise the percenttage of the capacity of hemapoietic tissue in mice with the radiation damage.
      Conclusion   The injection of acanthopanax senticosus can significantly repair the bone marrow microangium and showed a protective role on the bone marrow hematopoietic micro environment in mcie with the radiaiton damage.
