免疫-PCR早期诊断肾综合征出血热的研究

Study on immuno-PCR technique for early diagnosis of hemorrhagic fever with renal syndrome

      目的   建立一种敏感、特异的肾综合征出血热(HFRS)早期诊断方法。
      方法   应用免疫-PCR方法检测不同病程、临床诊断为HFRS的患者血中抗HFRS-IgM抗体, 同时与ELISA和IFAT二种方法进行比较。
      结果   免疫-PCR检测96份HFRS患者血清中抗HFRS-IgM抗体的阳性率为76%, 对51份发热期患者的阳性检出率为62.7%, 而ELISA法和IFAT法对同批患者的阳性检出率分别为57.3%和39.2%, 及49%和31.4%.20份甲型肝炎阳性血清、30份正常人血清及20份HFRS-IgM阴性并掺入类风湿因子阳性的血清, 免疫-PCR检测均为阴性。
      结论   免疫-PCR检测抗HFRS-IgM抗体敏感性高, 特异性强, 可作为HFRS早期诊断方法。

     

    Abstract:
      Objective   To seek a sensitive and specific method for early diagnosis of Hemorrhagic Fever with Renal Syndrome(HFRS).
      Methods   The 96 sera from patients with HFRS diagnosed by clinic were tested with immuno-PCR te-chonique for anti HF RS-IgM antibody and compared with ELISA and IFAT, and a control group(20 sera with HAV positive, 30 sera of normal human and 20 sera of HFRS IgM negative mixed with the sera of rheumatoid factor).
      Results   For the 96 sera, the positive rate with Immuno-PCR was 76% and among the 96 sera, 51 sera were from patients with febrilestag and the positive rate with the immuno-PCR was 62.7%.For the latter two methods, the positive rate were 57.3% and 39.2%, or 49% and 31.4% respectively.20 sera with HAV positive, 30 sera of nor mal human and 20 sera of HFRS IgM negative mixed with the sera of rheumatoid factor were all shown negative.
      Conclusion   The method of Immuno-PCR for detection of anti HFRS-IgM antibody was sensitive and specific, which may become a new method for early diagnosis of HFRS.
