烹调油烟对大鼠肺组织细胞周期和凋亡的影响

Effect of cooking oil fumes on cell cycle and apoptosis in lung cells of spraque dawley rats

      目的   检测SD(Spraque dawley)大鼠吸入烹调油烟(cooking oil fumes, COF)后肺组织细胞周期和凋亡的变化。
      方法   以SD大鼠作为实验对象, 通过动式吸入染毒, 于染毒后第11个月末, 采用流式细胞仪(flow cytometer, FCM)技术检测肺组织的细胞周期和凋亡。
      结果   G1期细胞所占百分数随染毒剂量的增加而减少, 各组间的G1%有显著性差异(F=5.08, P < 0.05); S期细胞所占百分数随染毒剂量的增加而增高, 组间S%有显著性差异(F=17.06, P < 0.01)。各组间肺组织细胞凋亡率有显著性差异(χ2=51.17, P < 0.01), 且存在剂量-反应关系(r=0.805, P < 0.05)。
      结论   在细胞周期中, G1期细胞所占百分数随染毒剂量的增加而减少, S期细胞所占百分数随染毒剂量的增加而增高; 肺组织细胞凋亡率随染毒剂量的增加而减少。

     

    Abstract:
      Objective   To detect the lung tissue cell cycle and apoptosis of spraque dawley(SD)rats after inhaling cooking oil fumes(COF).
      Methods   SD rats were used to make the chronic toxicity model.Exper iment groups were exposed to COF of three kinds of different concentration(6.88±0.31)、(15.06±0135)、(35.33±1.69)mg/m3 171 times in 11 months, 30 min per time.Control group had inhaled clean air.Flow cytometer technique was used to detect lung tissue cell cycle and apoptosis.
      Results   Lung tissue cell apoptosis rate decreased with the rise of COF concentr at ion.There was a significant difference among different dosage groups(χ2=51.17, P < 0.01)with dose-response relationship(r=0.805, P < 0.05).G1 cell percent decreased and S cell percent incr eased in cell cycle w ith the in COF concentration rise.There were significant difference among different dosage groups(F=5.08, P < 0.05;F=17.06, P < 0.01).
      Conclusion   G1 cell percentage decr eased and S cell percentage increased in cell cycle with the rise of COF concentration.The lung tissue cell apoptosis rate decreased with therising in COF concentration.
