Effect of nonylphenol (NP) on mRNA and protein expression of in ovarian cancer

      目的   观察环境雌激素化合物壬基酚(nonylphenol, NP)对卵巢癌细胞(PEO4)c-myc和p53mRNA及蛋白表达的影响, 以探讨NP雌激素活性的分子生物学作用机制。
      方法   将PEO4细胞在DMEM培养液中进行常规传代培养。采用MTT比色法观察NP对PEO4细胞增殖的影响, 用逆转录聚合酶链反应(RT-PCR)技术和免疫组化方法检测NP对c-myc原癌基因和p53抑癌基因表达的影响。
      结果   与溶剂对照组相比, 在(8~96)×10-7mol/L浓度范围内, NP与8×10-9mol/L雌二醇作用类似可显著促进PEO4细胞增殖(P < 0.05), 并表现出良好的剂量-效应和时间-效应关系。RT-PCR及免疫组化结果显示, 32×10-7mol/LNP可促进卵巢癌细胞PEO4c-myc和p53mRNA及蛋白表达。
      结论   NP具有雌激素样生物活性, 可促进雌激素反应性肿瘤细胞PEO4增殖, 这一效应主要是通过影响c-mycmRNA和蛋白质的表达而实现的。

     

    Abstract:
      Objective   To explore the effects of nonylphenol(NP)on mRNA and protein expression of c-myc and p53 in ovarian cancer PEO4 cells.
      Methods   Human ovarian cancer PEO4 cells were grown in DMEM medium.MTT assay was employed to investigate cell proliferation.RT-PCR and immunohistochemistry was performed to determine mRNA and protein expression of c-myc and p53.
      Results   Within the range of(8-96)×10-7 mol/L, NP stimulated proliferation in PEO4 cells with a dose-dependent and time-dependent fashion.RT-PCR and immunohistochemistry analyses revealed that treatment with 32×10-7 mol/L for 72 h resulted in increased mRNA and protein expression of c-myc and p53 with 2-3 fold.
      Conclusion   NP possessed estrogenic activity and could markedly stimulate proliferation in estrogen responsive ovarian cancer PEO4 cells.Up-regulation of c-myc mRNA and protein expression was suggested to be involved in this biological effect.
