诱导家蝇幼虫抗菌蛋白的提取及动力学研究

Extraction and dynamics study on antibacterial protein of housefly larva by inducements

  • 摘要:
      目的   比较两种方法提取家蝇幼虫抗菌蛋白抑菌活性, 并观察两种诱导源所诱导家蝇幼虫血淋巴中抗菌蛋白产生的动力学变化。
      方法   用金黄色葡萄球菌、大肠埃希菌诱导家蝇幼虫, 在诱导后12, 24, 36, 48, 60h收集家蝇幼虫, 80℃热水处死, 分别用血淋巴、组织匀浆提取抗菌蛋白后进行抗菌活性的测定。
      结果   用组织匀浆提取的抗菌蛋白抑菌效果明显好于血淋巴; 金黄色葡萄球菌、大肠埃希菌诱导家蝇幼虫产生抗菌物质达最大活性时间分别为诱导后48, 36h。
      结论   用组织匀浆提取家蝇幼虫抗菌蛋白是一种较好的方法; 诱导后36~48h的抗菌蛋白活性最大。

     

    Abstract:
      Objective   To conduct comparative studies on antibacterial activity of antibacterial protein from the induced housefly larva by two methods, and observation of dynamic changes of antibacterial protein.
      Methods   The housefly larva were injecteded by E.coli and S.aureus, compared with antibacterial activity of antibacterial protein from haemolymph and tissue homogenate after being induced 12, 24, 36, 48, 60 h.
      Results   Antibacterial protein from tissue homogenate was more antibacterial activity than that from haemolymph.After induced 48 h by S.aureus, antibacterial protein of housefly larva was more active against bacteria, but after induced 36 h by E.coli, antibacterial protein was more activity.
      Conclusion   Extration of antibacterial protein from tissue homogenate was a better method, and antibacterial protein was more activity after induced 36-48 h.
