全反式维甲酸与顺铂抑制肝癌细胞的研究

Study on inhibiting effect of ATRA and cisplatin on human hepatoma cell

  • 摘要:
      目的   研究全反式维甲酸、顺铂等对人肝癌细胞株(SMMC)-7721细胞端粒酶活性及细胞增殖的影响。
      方法   采用体外细胞培养法、细胞形态学检查、噻唑蓝(MTT)比色法、端粒酶重复扩增(TRAP)银染法。
      结果   全反式维甲酸可明显抑制SMMC-7721细胞端粒酶活性, 同时抑制肝癌细胞增殖, 细胞形态学出现恶性表型逆转; 顺铂不能抑制端粒酶活性, 但可抑制肝癌细胞增殖。
      结论   维甲酸可显著抑制端粒酶活性。

     

    Abstract:
      Objective   To investigate the mechanism of effect of A TRA and cisplatin on telomerase activity and proliferation of hepatoma.
      Methods   Cell culture, morphology MTT calorimetric assay and TRAP assay.
      Results   ATRA is the only one that can inhibit telomerasa activity, all agents can inhibit the proliferation of hepatoma, the inhibitory effect of cisplatin is the strongest.
      Conclusion   ATRA can inhibit obviously telomerase activity of hepatoma cells.
