Abstract:
Objective To explore the effects of fluoride on DNA damage and apoptosis in human embryo hepatocytes.
Methods DNA damage and apoptosis were measured after human embryo hepatocytes were incubated with 40, 80, and 160 Lg/ml sodium fluoride using Single Cell Electrophosis(SCGE)and Flow Cytometry(FCM), respectively.
Results The DNA damage of human embryo hepatocytes in the fluoride gruoup was significantly higher than the control group.Ridit value were 0.582, 0.815 and 0.892 respectively.The percentage of apoptosis in the middle and high fluoride group were markedly higher than the control group.Morevoer, the rate of DNA damage and the percentage of apoptosis in high fluoride group were significantly higher than the low fluoride group.
Conclusion Fluoride exposure can cause DNA damage and apoptosis in human embryo hepatocytes, and there existed dose-effect relationship to some extent.
