Abstract:
Objective To study hepatitis B virus(HBV)genotypes of patients with chronic hepatitis B in five cities of China.
Methods 18 typical full-genomic sequences, including A, B, C, D, E and F genotypes of HBV were analyzed and aligned.Universal outer primers and type-specific inner primers were designed on the basis of conserved nucleotide sequences from pre-S1 to S region of HBV.595 serum samples collected from patients with chronic hepatites B in 5 cities of China were tested by the PCR genotyping method, 4 samples genotyped by this method as A, B, C and coinfection of B and C were selected randomly and anlayzed by the cloning and sequencing.
Results Of 595 serum samples, 8(1.4%)were genotype A, 53(8.9%)genotype B, 360(60.5%)genotpye C 112(18.8%)were coinfection of genotype B and C 14(2.4%)A and C, 15(2.5%)A and B 6(1.0%)A、B and C, and remaining 27(4.5%)unspecified.Distribution of gentype B、C and coinfection of genotype B and C between 5 cities was significantly different(P < 0.05).The results of sequencing were identical with that genotyped by the PCR method.
Conclusion Genotype C was the most frequent genotype in patients with chronic hepatitis B in China, and genotype B was the next.Coinfections of two or more than two genotypes HBV were also found.Genotype distribution in patients with chronic hepatitis B in 5 cities studied was quite different.
