Relation between expression of ARmRNA in prostatic tissues and levels of sex hormones

  • 摘要:
      目的   研究雄激素受体mRNA的表达和性激素随增龄的变化, 探讨性激素与增龄对前列腺的影响。
      方法   通过放射免疫分析、核酸印迹分析(Dotblot), 检测性激素(睾酮、促黄体生成激素、皮质醇、雌二醇)的含量, 表达雄激素受体。
      结果   血清睾酮含量随增龄下降, 对照组(20~39岁)睾酮含量为(8.67±3.25)ng/ml, > 40岁各组均明显降低。血清促黄体生成激素含量随年龄增长而增高。血清皮质醇含量进行性增高, 40~49岁组增高尤为明显。血清雌二醇的含量以40~49岁组增高最为明显, > 50岁人群均波动于同一水平上。前列腺癌(CaP)杂交信号强于前列腺增生(BPH)与正常前列腺(NP)组织。
      结论   血清睾酮含量与年龄呈显著的负相关(r=-0.45, P < 0.0001), 促黄体生成激素的变化亦相反。皮质醇含量与年龄呈正相关。前列腺癌组织的ARmRNA高表达, 而良性前列腺增生的杂交信号与对照组接近。

     

    Abstract:
      Objective   To study the effect of AR and sex hormones as well as change with age on prostatic disease.
      Methods   AR was expressed by dot blot.Sex hormones contents(testosterone、LH、cortisone、estradiol)were determined by radioimmune method.
      Results   A progressive declines of serum testosterone levels and increase of serum LH levels with age were found.Serum cortisol contents were found increased with age.Serum estradiol levels in surveyed men over 40 were higher than that of the men in control group.The estradiol concentration did not change more in the men over 50.Dot blot: AR hybrid signal of prostate cancer was stronger than that prostate hyperplasia and prostate tissues.
      Conclusion   Testosterone levels was negatively correlated with age(r=-0.45, P < 0.000 1), Chang of LH was opposite.Cortisol contents was positively correlated with age.Analysis of dot blot showed that ARmRNA expression in CaP tissues was higher than that of controls, and that ARmRNA expression in BPH tissues was no difference compared with controls.
