顺铂与亚硒酸钠对淋巴细胞转化的影响

Effects of cisplatin and sodium selenite on transformation of human T lymphocytes

      目的   探讨亚硒酸钠对顺铂抑制人体T淋巴细胞转化作用的影响, 顺铂与亚硒酸钠对正常人体T细胞rRNA基因转录活性的影响及T细胞转化与rRNA基因表达活性之间的关系。
      方法   在细胞培养24h时加入顺铂0.05, 0.20, 0.50mg/L, 在细胞培养开始或在加入顺铂的同时加入亚硒酸钠0.05mg/L; 细胞培养72h后, 检测T细胞转化率及银染核仁组织区(Ag-NOR)数量的变化。
      结果   亚硒酸钠与不同剂量的顺铂共同处理淋巴细胞, 细胞转化率显著高于相应剂量顺铂单独处理者(P < 0.01~0.001), 并达到或超过阴性对照的水平。亚硒酸钠预先加入或与顺铂同时加入者无显著性差异。各组间Ag-NOR的表达频率以及各类随体联合均无显著性差异。
      结论   亚硒酸钠可消除顺铂对T细胞转化的抑制作用, 且亚硒酸钠对T细胞转化及对顺铂作用的影响无时间效应。顺铂与亚硒酸钠对人体正常T细胞rRNA基因表达的影响以及T细胞转化与rRNA基因转录活性之间的关系仍不清楚。

     

    Abstract:
      Objective   To research the effect of sodium selenite on the action of cisplatin inhibiting transformation of human T lymphocytes, explore the influence of cisplatin and sodium selenite on rRNA gene transcription activity of lymphocytes and the relationship between transformation of T cells and rRNA gene transcription activity.
      Methods   Cisplatin was administered to the culture of lymphocytes after incubation for 24 hours with three different dosages(0.05, 0.2, 0.5 mg/L), the culture was also supplemented with sodium selenite(0.05 mg/L)at the beginning of the cell culture or at the same time as cisplatin was administered for different trea-t ment.The slide specimens were prepared using cytogenetical method after the cells were cultured for 72 hours, and then, the changes of the rate of transformation of T lymphocytes and frequencies of Ag-NORs, which reflect rRNA gene transcription activity, were investigated.
      Results   The rates of T lymphocyte transformation in the groups coneated with sodium selenite and cisplatin were significantly increased(P < 0.01-0.001)as compared with the groups without sodium selenite supplementation and suffering exposures to cisplatin alone at corresponding dose, up to or over the rate of negative control group.There were no significallt differences in the ratios of lymphocyte transformation between the groups pretreated by sodium selenite and the groups cotreated by sodium selenite with cisplatin, on condition that the other treatments were the same.There were no remarkable differences(P > 0.05)in the frequencies of AgNOR and satellite association among the all groups.
      Conclusion   Sodium selenite supplementation can preclude the inhibition of tranformation of T lymphocytes in vitro by cisplatin, regardless of pretreatment by sodium selenite or cotreatment with cisplatin, it is not time-dependent for sodium selenite precluding the inhibition of transformation of T lymphocytes by cisplatin.It is not clear the inflence of cisplatin or/and sodium selenite on rRNA gene transcription activity in normal human T lymphocytes and the relationship between T lymphocyte transformation and rRNA gene transcription activity.
