不同地区福氏2a志贺菌质粒DNA多态性分析

Analysis on plasmid DNA polymorphism of S.flexneri 2a from different areas

      目的   对福氏2a志贺菌质粒DNA进行基因分型, 提高福氏2a志贺菌痢疾暴发流行时同源克隆的鉴定分析水平。
      方法   对93株分离自不同地区、不同时间的福氏2a志贺菌用质粒图谱和限制性内切酶图谱法进行基因分型和同源克隆鉴定。
      结果   质粒DNA分析表明, 不同地区、不同时间分离的93株福氏2a志贺菌具有13种不同的质粒DNA图谱特征(PLT1~PLT13型)和15种限制性内切酶图谱特征(ReT1~ReT15型)。
      结论   93株来自不同地区、分离自不同时间的福氏2a志贺菌分属于15个克隆群, PLT型和ReT型分布呈现多态性特征, PLT1/ReT1型是海南、江门、蚌埠、合肥、安庆地区福氏2a志贺菌的优势克隆群组。表型相同的福氏2a志贺菌实际上是遗传结构多态性群体的集合, 表型分析结果作为判断传染来源或传播途径的依据具有很大的局限性。

     

    Abstract:
      Objective   To identify homogenous clones of S1flexneri 2a, using gene distribution polymorphism of plasmid DNA of S.flexneri 2a.
      Methods   93 strains of S.flexneri 2a isolated from different areas at different time were detected by plasmid profiles and restriction endonuclease profiles to identify genetic type and homogenous clones of S.flexneri 2a.
      Results   Plasmid DNA analysis showed that the 93 strains of S.flexneri 2a isolated from different areas at different time possessed 13 different plasmid profile features (PLT1/-PLT13 type)and 15 different restriction endonuclease profile features(ReT1-ReT15 type).
      Conclusion   The 93 strains of S.flexneri 2a isolates obtained from different areas and at different time fell into 15 subclonial clusters.Although these isolates shared a common predominant clonial cluster(PLT1/ReT1 type)in Hainan, Jiangmen, Bengbu, Hefei, Anqing.The same phenotypingS.flexneri 2a was actually composed of various genetic polymorphism construction clonial clusters, there existed obvious difference in the clonial distribution and the population genetic construction.Phenotyping analysis for the determination of source of infection and the route of transmission will result in a one-side view.
