阿魏蘑菇提取物对肿瘤细胞p53表达的影响

Effect of pleurotes sapidus on expression of p53 protein expression of tumor cell

  • 摘要:
      目的   探讨新疆阿魏蘑菇提取物对体外培养的肿瘤细胞p53蛋白表达的影响, 从细胞凋亡角度研究其抗肿瘤机制。
      方法   采用肿瘤细胞体外培养技术及流式细胞技术, 观察阿魏蘑菇不同剂量的水提物及醇提物对体外培养的4种肿瘤细胞p53蛋白表达的影响。
      结果   MTT实验结果显示, 受试物对4种类型肿瘤细胞均具有较强的杀伤作用, 尤其以0.1g/ml剂量组为明显; HT实验结果表明, 经受试物处理后, 4种类型肿瘤细胞均观察到细胞核固缩, DNA浓缩并向核膜靠拢形成浓染致密颗粒, 并有典型凋亡小体出现。提示, 受试物可能通过诱导肿瘤细胞凋亡而发挥抗肿瘤作用; 流式细胞仪检测结果显示, 受试物处理后, 4种类型肿瘤细胞p53蛋白表达水平均有不同程度的上调, 尤其以醇提物作用24h后对Q3细胞p53蛋白表达水平的影响最为明显。
      结论   阿魏蘑菇提取物中的各组分可协同作用诱导肿瘤细胞促凋亡基因的表达, 从而达到抗肿瘤的目的, 有关阿魏蘑菇通过何种途径提高p53蛋白表达水平尚待深入探讨。

     

    Abstract:
      Objective   The anti-tumor mechanism of pleurotes sapidus on 4 kinds of tumor cell lines(Q3, MGC-803, Hela, SPC-A-1)was studied.
      Methods   Four tumor cell lines were cultured in vitro.The expression of p53 was investigated by FACS techniques after treated by water extract or ethanol extract from pleurotes sapidus with doses 10-1-10-3g/ml.
      Results   Experimental study of MTT and HT were used to determine the antitumor effects of pleurotas sapidus on inducing apoptosis of cancer cells.The incidence rates of apoptosis were remarkably increased after treated 12 hours and 24 hours by ethanol extract(0.1 g/ml)and water extract(0.1 g/ml).The effects of ethanol extract and water extract on expression of p53 gene related to apoptosis were examined by FACS technique.The expression of p53 protein in the groups of ethanol extract and water exract after treated 12 hours and 24 hours were significantly increased.
      Conclusion   The anti-tumor effect of pleurotas sapidus was evident, and its mechanisms was probably involved in the regulation of apoptosis by inducing the gene expressions relating to apoptosis, and up-adjustment improving apoptosis gene expression(p53).
