肿瘤血管生长因子抑制剂的研究

  • 摘要:
      目的   探讨低分子多糖对肿瘤血管生长因子的抑制作用。
      方法   血管生长因子抑制试验采用鸡胚绒毛尿囊膜法。体内抑瘤试验选用肉瘤S-180、肝癌H22、艾氏腹水癌等瘤株, 灌胃给药, 每天1次, 连续10d, 观察对实体瘤的抑制作用及对艾氏腹水癌的生命延长作用.体外细胞毒试验选用K562病、Hela宫颈癌细胞株, 采用体外细胞毒试验(MTT)法, 计算对肿瘤细胞株平均细胞生长抑制率及对细胞生长的抑制浓度。
      结果   低分子多糖能明显抑制鸡胚绒毛尿囊膜的肿瘤血管生成, 对肉瘤S-180的抑制率在31.7%~51.2%, 对肝癌H22的抑制率在30.1%~52.1%, 对艾氏腹水癌及肝癌H22腹水癌作用不明显。对K562白血病细胞株, 低分子多糖的半数抑制浓度IC50为20μg/ml, 对Hela宫颈癌细胞株的半数抑制浓度IC50为23.4μg/ml。
      结论   低分子多糖是较好的肿瘤血管生长因子抑制剂, 对肿瘤细胞株体内抑瘤试验、体外细胞毒试验均有明显的抑制作用。

     

    Abstract:
      Objective   To investigate the inhibitory effect of the low-molecular polysacchoride on tumor angiogensis factor(TAF).
      Methods   Chicken's embryonic chorio-allantois is adapted in the inhibitory experiment of TAF.In internal inhibitory experiment sarcoma line S-180 and cancer of the liver line H22 and EAC line are chosen.The medicine is poured down the stomach with one time per day for 10 days consistently.To observe the inhibitory effect of the medicine on sarcoma and the life extended effect of EAC line.In external cellular-poison experiment leukocythemia line K562 and Hela cervical carcinoma line are chosen.With the method of MTT, the average cellular-growth inhibitory rate and cellular-growth inhibitory concentration of cell lines are calculated.
      Results   The low-molecular polysacceharide inhibits TAF generation of the chicken's embryonic chorio-allantois obviously.The inhibitory rates of sarcoma line S-180 and cancer of the liver line H22 are 31.7%-52.1% and 30.1%-52.1% respectively.The effects on EAC line and cancer of the liver line H22 are not distinct.IC50 of the low-molecular polysaccharide is 20μg/ml for leukocythemia K562 and is 23.4μg/ml for Hela cervical carcinoma line.
      Conclusion   The low-molecular polysaccharide is a good inhibitor of TAF.The inhibitory effects of internal inhibitory experiment and external cellular-poison experiment on tumor cell lines are all very obvious.
