过量酒精对大鼠生精细胞凋亡的影响

Effect of ethanol on apoptosis in spermatogenic cells

      目的   研究酒精致生精细胞凋亡发生的机制及酒精凋亡相关基因Bcl-2、Bax在睾丸及生精细胞表达的影响。
      方法   利用人类饮用白酒制备大鼠的酒精毒性模型, 采用组织学技术和免疫组织化学技术检测酒精对生精细胞凋亡及其相关基因Bcl-2、Bax表达的影响.用2种剂量的酒精作用于大鼠的2个生精周期, 检测睾丸生精小管中Bcl-2、Bax蛋白表达的阳性细胞数和表达强度。
      结果   每个生精小管中Bcl-2蛋白表达的阳性细胞数在高剂量组为96±33.74, 低剂量组为156.6±28.52, 均显著低于正常对照组(222.6±56.86)(P < 001);每个细胞中Bcl-2蛋白表达强度(OD值)在高剂量组(0.142±0.035)、低剂量组(0.151±0.026), 都明显低于对照组(0.196±0.032)(P < 0.01);Bax蛋白表达的阳性细胞数在高剂量组为(412.4±96.75), 低剂量组为(337.5±94.39), 均明显高于对照组(2.14±82.57)(P < 0.01), Bax蛋白的表达强度(OD值)也有同样的变化, 高剂量组(0.113±0.036)和低剂量组(0.082±0.017), 均明显高于对照组(0.05±0.024)(P < 0.01).凋亡细胞增多, 平均每个生精小管中的凋亡细胞数目在高剂量组为15.53±3.75, 显著高于正常对照组(2.25±1.03)(P < 0.01)。
      结论   酒精可使生精细胞中Bcl-2的表达降低、Bax的表达增强, 生精细胞凋亡增加。

     

    Abstract:
      Objective   To study the effect of drinkingalcohol heavily on expression of Bcl-2 and Bax in spermatogenic cells.
      Methods   The animal model of drinking alcohol heavily was administered alcohol through gastric tube in rats.Then the expression of Bcl-2 and Bax in spermatogenic cells were investigated with the immunohistochemistry and image analysis.
      Results   The positive cell expression of bcl-2 in each genocyte was 96±33.74, 156.6±28.52 respectively, which was lower than the control(222.6±56.86, P < 0.01).Over doses use of alcohol for a long time could obviously reduce the number of cells of expressing Bcl-2 and the intensity of its expression(P < 0.01), and increase the number of cells of expressing Bax and the intensity of its ex pression in gonocyte(P < 0.01), increased the number of apoptosis of spermatogenic cells.
      Conclusion   Alcohol could reduced the expression of bcl-2, promoted the expression of Bax in gonocyte and the apoptosis of gonocyte.
