高效液相色谱法检测小麦、大米中赭曲霉毒素A

Determination on ochratoxin A(OA) in samples of wheat and rice by high performance liquid chromatography

  • 摘要:
      目的   建立赭曲霉毒素A的高效液相色谱检测方法; 了解沈阳地区小麦、大米中赭曲霉毒素A的污染状况。
      方法   利用液-液分离的前处理技术, 选用C18反相柱(250×4.6mm), 以乙腈: 0.008mol/L磷酸=56:44为流动相, 流速1ml/min荧光检测器(激发波长338nm, 发射波长455nm), 柱温30℃。
      结果   实验所得回收率和精密度以及确证试验的结果均令人满意。
      结论   建立了用高效液相色谱法检测赭曲霉毒素A的方法, 用本方法检测的62份小麦、大米样品中, 有1份小麦样品为阳性, 提示沈阳地区小麦和大米的赭曲霉毒素A污染率较低。

     

    Abstract:
      Objective   To build a method to deter mine OA in the sample with HPLC.
      Methods   Chromatography conditions: C18 column(250×416mm), mobile phase was acetonitrile 0.008 mol/L phosphoric acid(56:44, v/v), fluorescence detector(excitation wavelength 338 nm; emission wavelength 455 nm)termperature of column: 30℃.
      Results   The recovery rate and precision was satisfactory.
      Conclusion   This study established HPLC method to deter mine OA.One of sixty-two samples of wheat and rice was polluted by OA, which indicated contamination rate of cereal by OA was low.
