Effect of Fe on viability of heat-exposed K562cell and their hSP70 contents

  • 摘要:
      目的   观察微量元素铁对热暴露K562细胞活力及热应激蛋白HSP70影响, 探讨铁发挥热保护作用的可能机理。
      方法   以K562细胞为研究对象, 在普通培养基RPMI1640里分别添加了不同浓度的硫酸铁、柠檬酸铁配制成实验培养液, MTT法测定细胞活力, Tissieres方法测定热应激蛋白HSP70含量。
      结果   一定浓度的硫酸铁(100~400μmol/L)、柠檬酸铁(100~200μmol/L)能显着提高K562细胞的热耐力, 且热应激蛋白(HSP70)的合成也明显增加。
      结论   硫酸铁、柠檬酸铁能显着提高K562细胞的热耐力, 但它们作用浓度和效果不同, 其作用机理可能与热应激蛋白HSP70的合成增加有关。

     

    Abstract:
      Objective   To study the effects of Fe on viability of heat-stressed K562 cell, HSP 70 and its probable mechanisms.
      Methods   K562 cell were cultured with RPMI-1640 medium added with Fe at variant concentrations, then the cell viabilities of K562 and the contents HSP 70 were detected after heat exposure.
      Results   A certain concertration of Fe2SO4(100-400 μmol/L), critric acid iron(100-200 μmol/L)could enhance viabilities of heat-exposed K562 cell and their synthesis of HSP 70.
      Conclusions   These findings suggested that Fe could significantly promote thermoresistance of K562 cell, and some of their mechanisms might due to the enhancing synthesis of HSP 70.
