慈菇对镉致肝脏脂质过氧化及c-fos mRNA表达的影响

Effect of Sagittara Sagittifolia on changes of liver lipid peroxidation and expression of c-fos mRNA induced by cadmium

  • 摘要:
      目的   探讨慈菇对镉致肝脏脂质过氧化及c-fosmRNA表达的影响。
      方法   36只雄性SD大鼠随机分为6组(n=6), 分别用25%、50%、75%的慈菇(2ml/100g体重), 维生素E(50mg/100g体重)及等体积蒸馏水灌胃, 连续10d, 末次灌胃24h后, 腹腔注射CdCl 2(20μmol/kg体重), 染毒24h后, 比较各组肝脏丙二醛(MDA)、谷胱甘肽(GSH)含量及谷胱甘肽过氧化物酶(GSH-PX)活性; 并利用反转录聚合酶链式反应(RT-PCR)对肝脏c-fosmRNA进行半定量测定。
      结果   VE和中、高剂量慈菇都降低了镉所升高的MDA含量(P < 0.05), 中、高剂量的慈菇还提高了氯化镉所降低的肝脏GSH含量(P < 0.05);而VE和各剂量慈菇都不能纠正氯化镉所升高的c-fosmRNA水平(P > 0.05)。
      结论   镉对肝脏c-fosmRNA表达的诱导不是或仅小部分由脂质过氧化所介导。

     

    Abstract:
      Objective   To study the effect of liver lipid peroxidation on expression of c-fos mRNA induced by cadmium.
      Methods   The male Sprague-Dawley rats were randomly assigned into six groups, each of six.The rats were pretreated intragastrically with 25%, 50%, 75% Sagittaria Sagittifolia extract(2 ml/100 g°bw), vitamin E(50 mg/100 g·bw)or distilled water once daily for ten days.Twenty-four hrs after the last dosing, animals were administered with CdCl 2(20μmol/kg)intraperitoneally.Control animals were given distilled water only.Twenty-four hrs after the intoxication, the liver MDA, GSH content and GSH-PX activity were measured in each group above; liver c-fos mRNA was also semi-quantified by reverse-transcription polymerase chain reaction(RTPCR).
      Results   Pretreatment of rats with 5%and 75% Sagittaria Sagittifolia extract and vitamin E could decrease liver MDA content(P < 0.05), the former also increased liver GSH content(P < 0.05);However, both pretreatment of rats with Sagittaria Sagittifolia and vitamin E could not decrease liver c-fos mRNA levels induced by cadmium(P > 0.05).
      Conclusion   Lipid peroxidation seemed not or partially to involve in inducing expression of c-fos mRNA in the process of acute cadmium exposure.
