用RT-PCR方法直接检测麻疹病毒血凝糖蛋白(H)基因

Determination of measles virus hemagglutinin gene by RT-PCR

  • 摘要:
      目的   应用逆转录-聚合酶链反应(RT-PCR)法直接从临床麻疹患者的咽拭子及尿液中检出570bp的特异性H基因目的条带。
      方法   采集临床确诊为麻疹病例的咽拭子、尿液提取RNA, 用RT-PCR方法扩增麻疹病毒血凝糖蛋白的570bp核苷酸片段。
      结果   22例病人的20份咽拭子样品中, 均检测到相应条带, 20份尿液中有18份检测到相应条带; 采用RT-PCR一次扩增的产物, 检测灵敏度即达到0001TCID50。
      结论   麻疹病人唾液样本的RT-PCR阳性率高于尿液, 该方法可用于麻疹的病原学的快速辅助诊断, 特别是对出疹初期就诊的临床符合, 但麻疹IgM阴性的病例可进行进一步确诊。

     

    Abstract:
      Objective   Using RT-PCR to detect measles virus hemagglutinin gene(H gene)of 570bp length directly from throat swabs and urine of the measles patients.
      Methods   Extracting RNA from throat swabs and urine of the measles patients, using RT-PCR amplification measles virus hemagglutinin gene of 570bp.
      Results   Among 20 throat swabs of the 22 patients all were dedected the region.Among the 20 urine, 18 samples were dedected.By first PCR amplification, the minimum quantity of measles virus required to give a positive signal was < 0.001 TCID50.
      Conclusion   RT-PCR positive rate of the throat swabs was higier than the urine of measles patients.The method may help diagnose measles cases rapidly, espacially to those patients with rash in the initial clinical stage but with negative measles IgM.
