Study on arsenic toleranc induced by bone marrow mesenchymal stem cells

  • 摘要:
      目的   利用低剂量NaAsO2诱导骨髓间充质干细胞MSCs, 获得抗砷细胞, 探讨抗砷机制.
      方法   用1mol/L NaAsO2对MSCs进行低剂量慢性诱导, 利用四甲基偶氮噻唑蓝法(MTT)计算细胞生存率及半数致死量, 观察细胞抗砷性的产生.
      结果   NaAsO2诱导18周后, 人骨髓MSCs产生稳定抗砷性, 砷诱导组细胞LC50为25.8mol/L, 同步对照组细胞LC50为11.4mol/L, 砷诱导组细胞LC50是同步对照组细胞LC50的2.2倍.
      结论   低剂量NaAsO2长期诱导人骨髓MSCs, 可使细胞获得抗砷性.

     

    Abstract:
      Objective   To obtain stable arsenic tolerance in bone marrow mesenchymal stem cells(MSCs).
      Methods   Bone marrow mesenchymal stem cells were exposed atlow level arsenite.The cell surviral rate and LC50 were measured by MTT.The resistance to arsenite was observed.
      Results   Bone mar rowmesenchymal stem cells exposed to 1mol/L sodium arsenite for 18 weeks exhibited dramatic resistance to acute arsenite toxicity.The LC50 for acute arsenite exposure in these chronic arsenic-esposed cells was 25.8mol/L versus 11.4mol/L in control cells.
      Conclusion   Acquisition of stable arsenic toler ance in bone mar row mesenchmal stem cells was available.
