芥子气对淋巴细胞DNA损伤的研究

Study on DNA Damage in Lymphocytes Induced by Mustard Gas Intoxication

  • 摘要: 分离大鼠脾淋巴细胞培养24小时后,经10-1000μm芥子气染毒30min继续培养3d以单细胞凝胶电泳法检测DNA损伍结果显示中毒后淋巴细胞DNA迁移率和受损细胞百分率呈时间和剂量依赖关系;中毒4h高、中剂量与对照组相比呈显著性差异(p<0.01)提示单细胞凝胶电泳法检测DNA损伤用作评价芥子气细胞毒性指标的可能性。

     

    Abstract: Isolated spleen lyrnphocry tes of rat having been exposed to 10-1000μrn mustaid gas at 37℃ 30minutes DNA damage lymphocytes at 424 and 72h postexposureweremeasured with SCG assay T he results not only show ed that time-dependent relationship and dose dependent relationship between mustard gas and DNA m igration and the percentage of DNA-danaged lymphocytes but also a highly significant increase in DNA dan age at 10-3M,10-4M after 4h postexposure compared w ith the control group It's suggested that the method may be used as and indicator of crytotoxi;ity resulted fmmmustard gad and also it may play certain role in early diagnosis in mustard gas intoxication.
