应用小鼠胚胎肢芽培养方法研究重铬酸钾的发育毒性

The Utilization of mouse limb bud culture for evaluating the developmental toxicity of potassium dichromate in vitro

  • 摘要: 采用小鼠胚胎前肢芽培养方法研究重铬酸钾对小鼠的发育毒性。结果表明:重铬酸钾浓度在2.0mg.L-1 以上时,前肢芽器官的发育和分化受到影响,并且随着重铬酸钾浓度的增加,肢体中各软骨的发育分化越差,Neubert 评分得分越少,呈剂量- 效应关系。说明重铬酸钾为小鼠胚胎发育毒性物质。

     

    Abstract: Forelimb buds from 12th day mouse embryos were dissected and cultured in 0.5,1.0,2.0 and.410mg K2Cr2O7/LMEM+10%rat serum cultur ed for 6 days. The mouse forelimb development of the 0.5 and 1.0 mg. L- 1 groups were not significantly differ ent fr om the contr ols wher eas the higher concentr at ions resulted in a concentration related deterior at ion in limb developmen and differ entiation. The results indicated that potassium dichromate can inhibit mouse limb bud development and differ entiation in vitro. It is a potential developmental to xic substance in mice.
