快速、高敏感度检测Ki-ras基因突变方法的建立及其在检测大肠癌中的应用

  • 摘要: 建立一种检测Ki-ras基因突变的快速?高敏感度的方法,并将其应用于大肠癌的检测中?应用引物诱导限制富集突变等位基因检测法(PCR-PIREMA,PrimerIntroducedRestrictionwithEn-richmentofMutantAleleAssay)的敏感度达10-5~10-6,并在30例大肠癌样品中检测出了8例Ki-ras基因12密码子突变的样本,突变率达26.7%?证明RCR-PIREMA法是一种快速?敏感度高的方法,适用于在大量野生型拷贝中检测稀有的突变,可用于已知点突变的肿瘤的早期诊断及基因突变机制的动力学研究等许多方面?

     

    Abstract: To develop a rapid, highly-sensitive assay method to detect rare mutation of Ki-ras g ene, and to apply it in the detection of colorectal tumors. Methods By using an assay method named PCR-PIREM A( Primer Introduced Rest riction w ith Enrichment of Mutant Allele Assay), thirty samples of colorectal tumors w ere analyzed. Results Eight ( 26. 7% ) of thirty contained mutations. The sensitivity of this method is confirmed to be 10 - 5 ~ 10 - 6 through the reconst ruction experiments. Conclusion PCR-PIREM A is a rapid, highly-sensitive assay. It is necessary for early detection of cancers with highly f requency of known point mutations and kinetics studies of mutagenesis induced by carcinogens at particular bases of a gene.
