氟对大鼠骨组织Osterix mRNA及其蛋白表达影响

Effect of fluoride on expression of OSX mRNA and protein in bone tissue of rats

  • 摘要: 目的 观察氟对大鼠股骨中成骨细胞特异性转录因子Osterix(OSX)mRNA及其蛋白表达的影响。方法 36只SD大鼠按体重随机分为对照组、低氟组(饮水含氟50 mg/L)、高氟组(饮水含氟5 mg/L),饲养6个月后股动脉放血处死。用免疫组织化学法检测各组大鼠股骨组织中OSX蛋白表达;实时荧光定量PCR检测OSX mRNA表达。结果 高氟组大鼠OSX阳性成骨细胞数(51.47±3.37)个增多,明显高于低氟组(36.80±2.83)个和对照组(27.00±2.92)个,差异有统计学意义(P<0.05);低氟组OSX mRNA是对照组的2.10倍,高氟组OSX mRNA是对照组的2.82倍,与对照组比较,差异有统计学意义(P<0.05)。结论 氟可导致大鼠骨组织OSX mRNA及蛋白表达水平增高,OSX可能参与氟引起的骨骼损伤的发生机制。

     

    Abstract: Objective To investigate the changes of mRNA and protein expressions of osterix(OSX)in the bone of rats with chronic fluorosis. Methods Thirty-six SD rats were randomly divided into three groups(12 in each group with half male and half female):control group,low and high fluoride group.The fluoride groups exposed to sodium fluoride (low:NaF of 5 mg/L and high:NaF of 50 mg/L,respectively)added to the drinking water to produce the animal model of fluorosis.The expressions of OSX protein and OSX mRNA inside the distal femur of the rats with chronic fluorosis were detected by immunohistochemical technique and real-time quantitative PCR,respectively. Results Compared with the control group,mRNA and protein expressions of OSX were increased in low and high fluoride rats,the difference was statistically significant(P<0.05). Conclusion Fluoride might induce the mRNA and protein expressions of OSX,and the change of OSX may be involved in the increase of the bone turnover and be one of the the pathogenetic factors in fluorosis.
