白藜芦醇甙对HIBD新生大鼠皮层BDNF表达影响

Effects of polydatin on expression of BDNF in cortex of neonatal rats with hypoxic-ischemic brain damage

  • 摘要: 目的了解白藜芦醇甙(PD)对新生大鼠缺氧缺血性脑损伤(HIBD)大脑皮层脑源性神经营养因子(BD-NF)表达的影响。方法 52只新生7日龄SD大鼠随机分为假手术对照组(NS组)、阴性对照组(NG组)、缺氧缺血组(HI组)、白藜芦醇甙干预组(PD组);通过结扎左颈总动脉及通入8%的氧气2 h制备新生大鼠缺氧缺血性脑损伤模型;各组按照造模后24 h、10 d分为2个亚组,免疫组化法测定各组大鼠大脑皮层BDNF的表达。结果免疫组化结果显示24 h、10 d HI组大鼠皮层BDNF表达(0.144±0.011,0.105±0.011)较NS(0.069±0.005,0.068±0.005)和NG组(0.070±0.003,0.069±0.007)增高(P<0.01),10 d PD组皮层BDNF表达(0.131±0.009)较HI组(0.105±0.011)明显增加(P<0.01)。结论白藜芦醇甙能上调HIBD新生大鼠大脑皮层BDNF的表达,对缺氧缺血后的神经元有保护作用。

     

    Abstract: ObjectiveTo investigate the protective effects and possible mechanisms of polydatin on the expression of brain-derived neurotrophic factor(BDNF)in cortex of neonatal rats with hypoxic-ischemic brain damage(HIBD).MethodsFifty-two neonatal SD rats were divided randomly into normal sham-operated(NS),negative control(NG),HIBD(HI), and polydatin treatment group(PD).The HIBD model was established in 7 days old rats by left carotid artery ligation and 2 hours hypoxia.Two subgroups were individed according to the model after 24 hours and 10 days.Immunohistochemistry was used to measure the expression of BDNF in cortex.ResultsThe results of immunohistochemistry demonstrated that the expressions of BDNF in cortex in HI groups at 24 hours and 10 days(0.144±0.011,0.105±0.011)were significantly higher than those in NS group(0.069±0.005,0.068±0.005)and NG group(0.070±0.003,0.069±0.007)(P<0.01 for all);the expression of BDNF in cortex in PD group(0.131±0.009)was significantly higher than that in HI group (0.105±0.011)(P<0.01).ConclusionPolydatin had possible neuroprotection effect in hypoxic-ischemia brain injury and the mechanism was probably related to the upregulation of the expression of BDNF in cortex in neonatal rats.
