丙泊酚对类风湿性关节炎大鼠炎症反应及滑膜细胞凋亡影响

Effect of propofol on inflammation and synoviocyte apoptosis in rheumatoid arthritis rats

      目的  探讨丙泊酚对类风湿性关节炎(RA)大鼠炎症反应及滑膜细胞凋亡的影响。
      方法  建立佐剂诱导关节炎(AIA)大鼠模型并用丙泊酚(2.5、5.0、10.0 mg/kg)处理,HE染色观察关节病变,ELISA检测血清中肿瘤坏死因子 α(TNF-α)、白介素1β(IL-1β)和IL-6浓度。蛋白质印迹检测葡萄糖调节蛋白(GRP78)、生长停滞与DNA损害诱导蛋白(GADD34)、C/EBP同源蛋白(CHOP)、核因子(NF-κB p65)及NF-κB抑制蛋白(IκBα)在大鼠滑膜组织中的表达。
      结果  丙泊酚可减小AIA大鼠足趾体积,减轻关节组织炎性浸润及滑膜细胞增生,降低TNF-α、IL-1β 和IL-6在AIA大鼠血清中的浓度;抑制AIA大鼠关节炎滑膜组织中GRP78、GADD34、p-IκBα 和NF-κB p65表达,促进CHOP表达。
      结论  丙泊酚可抑制类风湿性关节炎(RA)大鼠炎症反应,其机制可能与调控内质网应激有关。

     

    Abstract:
      Objective  To investigate the effect of propofol on inflammation and synoviocyte apoptosis in rheumatoid arthritis rats.
      Methods  Adjuvant-induced arthritis (AIA) rat model was established and then the AIA rats were administered with propofol at dosages of 2.5, 5, and 10 mg/kg. Pathologic changes in joints of the rats were observed using hematoxylin-eosin (HE) staining; serum concentration of interleukin-1 beta (IL-1β) and interleukin-6 (IL-6) were measured with enzyme-linked immunosorbent assay (ELISA). The expressions of 78-kD glucose-regulated protein (GRP78), growth arrest and DNA damage inducible gene 34 (GADD34), CCAAT/enhancer-binding protein (C/EBP) C/EBP homologous protein (CHOP), nuclear factor-κB (NF-κB) p65 and inhibitor of NF-κB alpha (IκB α) in the rats′ synoviums were determined with Western blot.
      Results  We observed diminished size of toes, decreased inflammatory cell infiltration and synovial hyperplasia in the synovial tissue, reduced serum concentration of TNF-α, IL-1β and IL-6, inhibited the expressions of GRP78, GADD34, p-IκBα and NF-κB p65, and promoted expression of CHOP in the AIA rats exposed to propofol.
      Conclusion  Propofol could inhibit the inflammation possibly through regulating endoplasmic reticulum stress in rats with rheumatoid arthritis.
