溴氰菊酯对大鼠脑中EROD酶活性及蛋白表达的影响

Effect of Deltamethrin on EROD activity and protein expression in rat brain

  • 摘要: 采用荧光分光光度法,在体内、体外研究了溴氰菊酯对大鼠脑组织中EROD(7-ethoxyresorufin O-deethylase)酶活性的影响.结果表明:大鼠经溴氰菊酯12.5mg·kg-1·d-1通过腹腔注射连续5天后,溴氰菊酯明显抑制全脑和大脑皮层EROD酶活性,抑制率分别是32.0%,37.0%;在体外,溴氰菊酯浓度在2×10-8~2×10-4mol/L范围内对EROD活性无明显影响;进一步用蛋白印迹技术检测CYP1A1/1A2蛋白表达的变化,结果表明:溴氰菊酯在体内明显抑制上述各脑区中CYP1A1/1A2蛋白表达.综合上述两方面结果,提示溴氰菊酯在体内能明显抑制脑内EROD活性,其机理与抑制该酶蛋白合成有关.

     

    Abstract: The EROD activity in rat brain was determined by flurophotometry under the treatment of DM in vivo and vitro.The results showed:in vivo,DM could markedly inhibit EROD activity in rat brain microsome after rats had been received DM(12.5mg·kg-1·d-1,ip)for 5 days.The inhibitory rate in whole brain,cerebral cortex were 32.0% and 37.7% respectively.However,in vitro, under the concentration of 2×10-8~2×10-4mol/L,there was no obvious change of EROD activity in rat brain.Moreover,Western blot analysis indicated that DM could significantly reduce the expression of C YP1A1/1A2 protein.So the mechanism by which DM inhibit EROD activity in rat brain was releated to the reduction of C YP1A1/1A2 protein expression by DM.
