热休克蛋白-抗原肽复合物对移植黑色素瘤B16细胞小鼠的抑瘤效应

Inhibiting tumor effects of heat shock protein antigen peptide complex on mice with transplanted melanoma B16 cells

  • 摘要: 制备黑色素瘤B16细胞热休克蛋白抗原肽复合物(HACs)及其粗提物(HACCEs),并进行其抑瘤效应的研究.结果表明应用凝胶过滤制备的HACCE3、HACCE4和HACCE5及应用亲和层析纯化的HAC60、HAC75和HAC97均不同程度地降低肿瘤发生率、延迟肿瘤发生时间和减少C57BL/6J小鼠死亡率.

     

    Abstract: In the present study,we prepared heat shock protein-antigen peptide complexes(HACs)and their crude extracts(HAC-CEs) from melanoma B16 Cell line,and studied on their inhibiting tumor effects.HAC2CE3 CE4 and HAC2CE5 prepared with gel filtration and HAC60,HAC75 and HAC97 purified with affinity chromatograhy all decreased the incidences of tumors,delayed the times of tumor development and reduced the mortality of C57BL/6J mice to varying degrees.The results suggest that HACs of 60~97 kD have the immunogenicity and the effects of inhibiting tumor,and offer the important experimental evidences for preparing tumor vaccines.
