出血性大肠杆菌O157:H7DNA文库的构建

The Construction of Enterohaemorrhagic E.Coli O157:H7 DNA Bank

  • 摘要: 目的为建立出血性大肠杆菌O157:H7DNA文库,筛选致病岛LEE.方法提取EHECO157:H7EDL933染色体,Xmal部分酶切并与pJB8X连接、包装,感染E.coliDH5α,鉴定库容.结果库容约700克隆子,大于608个理论值,构建成功.结论成功构建了出血性大肠杆菌O157:H7DNA文库,为目的片段筛选打下良好基础.

     

    Abstract: ObjectivesTo construct EHEC O157:H7 DNA bank forth screening of patho genicity island LEE.MethodsChromosomal DNA of Enterohaemorrhagic E.Coli O157:H7 was extracted and digested with Xmal.Then it was ligated to pJB8X and packed with Kphages.The products of package were used to infect E.coli DH5α and the capacity of DNA bank was identified.ResultsThe content of the bank was about 700 clones,which was larger than that of the theo retical value.ConclusionThe bank has been constructed successfully for screening any fragment you want.
