大肠杆菌O157∶H7在8种水质中存活力观察

Investigation of the Survial of Escherichia Coli O157∶H7 in Eight Kinds of Water Quality

  • 摘要: 目的 探讨EHECO157:H7在各种水质中存活力.方法 采集8种不同水质的水样,将适当浓度的第5代EHCEO157:H7933株菌悬液加入水样中,使染菌浓度为106CFU/ml,染菌后每隔5天采样1次,进行细菌的生化鉴定和血清学鉴定确认后予以计数.结果 EHECO157:H7污染水样后存活时间最短的为15天,最长的达40天以上;EHECO157:H7在4℃游泳池新水、矿泉水中存活量呈急剧衰减,在4℃井水中存活量则在15天内剧降2个数量级后略呈平稳接近104CFU/ml之后又快速衰减,在其他水样4℃条件下存活数量变化均呈逐渐衰减,降至103CFU/ml~104CFU/ml数量级(池塘水位于105CFU/ml数量级)后存活菌量急剧减少;EHECO157:H7在25℃井水中存活量5天内急剧衰减3个数量级后在103CFU/ml呈平稳持续至第15天后衰减再次加速,在25℃纯净水逐渐衰减,在其余25℃水样中则均呈急剧衰减.除在25℃游泳池旧水中第10天时其存活量高于4℃游泳池旧水外,在其余水样25℃条件下EHECO157:H7存活量均比同期4℃条件下水样少,而且在25℃水样中存活菌量衰减速度明显较快.结论 EHECO157:H7在水中存活情况与水质种类、水温以及水中有机物、抑菌物质含量密切相关;在EHECO157:H7水型暴发疫情控制中,

     

    Abstract: Objective To investigate the survival of EHEC O157:H7 in various kinds of water quality.Methods We collected eight kinds of water sapmples and added the solution of the fifth generation of EHEC O157:H7 933 strain bacteria suspended with appropriate concentration to the water samples which had been sorted in the laboratory and made the concentration of bacteria infected up to 106 CFU/ml,then preserved these water samples under the condition of 4℃ and 25℃ individually.Samples were collected once every five days after infecting bacteria,then the bacteria were separated and identified.The bacteria were sifted primarily through SMAC、disaccharidferric、lysine dynamic agar before separation and identification.The Colonys whose feature coincided with EHEC O157∶H7 were counted after the assurance of bacteria biochemical identification and serum identification.Results The survial time of EHEC O157:H7 lasted 15 days at least and 40 days at most after they polluted water samples;the Survival number of EHEC O157:H7 indicated acute attenuation curve in swimming-pool new water or mineral water under 4℃ and the survival number slightly indicated the plateau near 104CFU/ml after it decreased by 2 orders of magnitude acutely in 15 days in well-water under 4℃ and the survival number's change of EHEC O157:H7 indicated double-plateau attenuation curve in other samples under 4℃;The survival numberof EHEC O157:H7 in well-water under 25℃ indicated plateau's another acute attenuation after it decreased by 3 orders of magnitude acutely in 5 days;The survival numbers all indicated acute attenuation curves under 25℃ in other water samples;The survival numbers of EHEC O157:H7 under 25℃ in other water samples were all fewer than in the same time's samples under 4℃ except that their survival numbers were higher in swimming-pool old water on the tenth day than in that under 4℃;The survival numbers of EHEC O157:H7 all indicated the phenomenon of attenuation's sudden acceleration when they decreased to 104CFU/ml in various kinds of water samples under different temperatures except in well2water under 25℃.Conclusion The survival of EHEC O157:H7 in water quality is associated tightly with the kinds of water quality、surrounding's temperature、organic substance in water and content of bacteriostatic substance;Attention should been paid to the survival difference of this kind of bacterium when searching source of infection and dealing with infectious water,then different measurements are adopted; No drinking fresh water is advocated in daily preventive work and supervision and monitoring should been strengthened in the production of pure water and mineral water;Water in swimming-pool should be changed and swimming-pool be sterilized on the stipulation.
