原发性肝细胞肝癌细胞凋亡与bcl-2基因的关系

The Relationship between Apoptosis and bcl-2 Gene in Primary Hepatocarcinoma

  • 摘要: 目的 研究细胞凋亡相关基因bcl-2与原发性肝细胞肝癌(HCC)细胞凋亡的关系.方法 采用TdT酶介导的生物素化dutp缺口末端标记技术(Tunel)和免疫组化SP法,检测肝癌组织的细胞凋亡指数和bcl-2基因的表达并以慢性肝炎肝组织作为对照.结果 慢性肝炎肝组织细胞凋亡指数显著高于癌组织细胞(P<0.01),bcl-2在癌组织中的表达为51.3%,在对照组中的表达为25%.肝癌组织中bcl-2表达与癌组织凋亡指数呈负相关.结论 在HCC中,癌细胞凋亡明显受到抑制,其发生可能与bcl-2基因调节障碍有关.

     

    Abstract: Objective To study the relationship between apoptosis and apoptosis-related gene bcl-2.Methods Using terminal deoxynuleotidyl transferase mediated dUTP biotin nick end labeling(Tunel)and immunohisto chemistry SP method,the apoptosis rate and the expression of bcl-2 in HCC was examined.Results The apoptosis rate was significantly higher in chronic hepatitis than in HCC(P<0.01).The frequency of expression of bcl-2 in HCC was 51.3%,while that in chronic hepatitis was 25%.The expression of bcl-2 was negatively correlated with the apoptosis(P<0.01).Conclusion Apoptosis is inhibited in HCC which may be related to bcl-2 gene dysregulation.
