应用PCR/SSCP分析恙虫病东方体分离株基因型的研究

Study on Gene Type of Orientia Tsutsugamushi Isolated Strains by PCR/SSCP

摘要: 目的对分离自辽宁、吉林地区的6株恙虫病东方体目的基因进行分析研究,并与国际参考株进行比较.方法经PCR/SSCP检测,呈现出2种不同的单链构象图谱:HCAa9202株、HCAp9203株及HCM9501株图谱与Gilliam参考株相同,而KDCt9402株、KDCp9403株及KDCt9404株的图谱则与Karp参考株相同.结果辽宁、吉林地区恙虫病东方体分离株至少存在2种型别.结论应用PCR/SSCP方法分析恙虫病东方体基因型,简便、经济、省时、结果准确.

Abstract: ObjectiveUsing PCR/SSCP to analyse the gene type of the six Orientia stustugamushi(Ot)strains isolated from Liaoning and Jilin areas and making a comparision between the isolated strains of Ot and the international reference strains.MethodsThe six strains from Liaoning and Jilin appeared two different kinds spectrum of SSCP:The strains HCAa9202,HCAp9203 and HCM 9501 had the same gene type with the reference strain Gilliam;strains KDCt9402,KDCp9403 and KDCt9404 had the same gene type with reference strain Karp.ResultOt strains isolated from Liaoning and Jilin areas had at least two different types identified by PCR/SSCP analysis.ConclusionThe method of PCR/SSCP was proved to be simple,economic,time-saving and accurate.
