人源化抗CD3单链抗体的构建、序列分析和表达研究

Study on Construction and Expression of Anti-CD3 Single Chain Fv Gene and DNA Sequence Analysis

  • 摘要: 目的构建和表达抗CD3单链抗体,并对其构象进行模拟分析.方法应用重叠延伸拼接法连接抗CD3抗体的轻链可变区基因和重链可变区基因,构建抗CD3单链抗体基因(ScFv),并将其克隆于原核表达载体中表达.结果拼接后的抗CD3ScFv基因全长732bp,构象模拟分析表明其三维结构稳定,兼容性分数S>0,表达的目的蛋白约26.5kD.结论抗CD3ScFv基因的成功构建,为进一步研究抗CD3/抗乙肝病毒的双特异性抗体奠定了基础.

     

    Abstract: ObjectiveTo construct and express the single chain Fv gene fragment for human CD3 antigen and conduct imitation analysis on conformation.MethodsThe variable region gene of heavy chain(VH)and light chain(VK)of antiCD3 antibody were linked into single chain Fv by splicing overlapping and extension(SOE)ScFv genes were cloned to pET28(a+)and expressed in E.coli.ResultsThe DNA sequence analyses and imitation of conformration analyses demonstrated that ScFv gene were 732bp,conformation steady.compatibility S>0.The ScFv fragment were expressed in BL21(DE3),and folded into 26.5 KD protein.ConclusionThe correct gene of anti-CD3 Scfv was constructed.It would be used to construct the anti-CD3/anti-HBV bifunctional antibody.
