二阶导数光谱法测定华法林

Using Second-Derivative Ultraviolet Spectrometry to Determine Warfarin

  • 摘要: 目的 介绍一种应用二阶导数紫外光谱测定尿和血浆中华法林的新方法.方法 尿和血浆用甲醇/醋酸缓冲液稀释后,通过PT-C18柱进行固相萃取,用氯仿洗脱.洗脱液用NaOH溶液反提,测定NaOH溶液的二阶导数光谱,用328nm处的吸光度值时行定量.结果 尿中华法林的检测下限为05~1mg/L,血浆则为2~4mg/L.结论 该方法可用于大剂量中毒检测,亦可用于微量血药浓度检测.

     

    Abstract: Objective To introduce a new method using a second-derivative ultraviolet(UV)spectrometry to determine warfarin in human urine and plasma.Methods The drug-containing samples,after mixing with methanol acetate buffer,were directly applied to the cartridges and eluted with chloroform.The eluate was strip-extracted with NaOH solution.The stripping solution was subjected to UV analysis with the absorption peak at 328nm in the second-derivative spectr a.Results The detection limits were 0.5-1.0μg·ml-1 forurine and 2-4μg·ml-1 for plasma,respectively.Conclusion This method can be used in the detection of big-dosage poision and trace drug content in blood.
