应用载体法观察甲醛熏蒸HBsAg的破坏作用

Study On Efficacy of Formaldehyde Fumigation Destroying HBsAg By Carriers Test

  • 摘要: 目的 以乙型肝炎表面抗原(HBsAg)作为指标,评价甲醛气体对乙型肝炎病毒(HBV)的灭活作用.方法 用直径为1cm的原型钢片、乳胶片和1.5cm×1.5cm麻布片制备3种载体片,在每个载体片上滴加含纯化HBsAg2μg的悬液.在熏蒸橱内4角和中间放置载体片,用高锰酸钾氧化多聚甲醛产生甲醛气体熏蒸,在含1%小牛血清的磷酸盐缓冲液中加入0.5%双甲酮和0.3%吗啉为回收液,用放射免疫法(SPRIA)检测HBsAg.结果 在温度为25℃、湿度为75%、消毒时间为2h条件下,随多聚甲醛消毒剂量的增加,消毒效果增强.多聚甲醛剂量为15g/m3时,可将乳胶、布片载体上HBsAg破坏(S/N≤2.1),达到20g/m3时,可将3种载体上的HBsAg破坏.在相同的温湿度条件下,多聚甲醛消毒剂量为10g/m3时,随消毒时间的延长,消毒效果增强;达到5h时,可将乳胶、布片载体上的HBsAg破坏,对钢片载体上的HBsAg达不到消毒效果(S/N>2.1).结论 多聚甲醛经用高锰酸钾氧化产生的甲醛气体在一定温湿度条件下,经一定消毒时间,对HBsAg有较好的破坏作用,物品表面粗糙比表面光滑消毒效果好.

     

    Abstract: Objective To evaluate the efficacy of formaldehyde fumigation destroying HBV taking HBsAg as index.Methods Three kinds of carriers (stainless steel,emulsion round plates with diameter of 1.0cm,and cloth square plates with 1.5×1.5cm2) were prepared.20 μl of HBsAg suspension (100μg/ml) containing 1% calf serum with phosphate buffer was droped on each carrier.The carriers were dried in an incubalor (37℃) for 2 hours,then kept in refrigerator (4℃) before used.Formaldehyde fumigation was conducted in the fumigator HBsAg was tested by SPRIA.Results The efficacy of fomlalde-hyde fumigation destroying HBsAg was directly proportional to the paraformaldehyde dose under the conditions of sterilizing 2 hours,temperature 25℃,relative humidity 75%.While paraformaldehyde dose reached 20g/m3,the HBsAg on all 3 kinds of carriers was destroyed.The efficacy of formaldehyde fumigation destroying HBsAg was directly proportional to the sterilizing hours(under the conditions of paraformaldehyde dose 10g/m3,temperature 25℃,relative humidity 75% ).While formaldehyde fumigation hours reached 5 hours,the HBsAg on the emulsion and cloth plates was destroyed,but on stainless steel plates was not.Conclusion It was suggested that the rough surface of material retain more fomlaldehyde gas and the HBsAg on it was destroyed easily,the smooth surface of material retain less formaldehyde gas and the HBsAg on it was destroyed difficultly.
