氯化镉诱导293细胞凋亡的实验研究

Study on Apotosis of 293 Cells Induced by Cadmium Chloride

  • 摘要: 目的 采取体外细胞培养方法观察CdCl2在不同时间、剂量作用下,诱导293细胞凋亡关系.方法 实验中选择0,12.50,25.0,50.0μmol/L共4种浓度CdCl2在体外与293细胞共育不同时间后(12h,24h,48h),采用台盼蓝拒染法测定细胞存活率,观察293细胞转化,用DNA凝胶电泳及ELISA方法检测细胞凋亡.结果 12.5,25.00,50.0μmol/L CdCl2可诱导细胞凋亡;不同浓度CdCl2可引起细胞存活率下降,与CdCl2时间、剂量有关.结论 CdCl2在体外可诱导293细胞凋亡,与CdCl2的作用时间、作用浓度相关.

     

    Abstract: Objective To explain apoptosis induced by cadmium chloride in 293 cells in vitro.Methods 293 cells were exposed to 0,12.5,25.0,50.0μmol/L of CdCl2 for various time period,to measure the apoptosis by DNA agarose electrophoresis and ELISA.Results CdCl2 could induce apotosis of 293 cells in vitro at concentration of 12.50,25.00,50.00μmol/L.Different concentration of CdCl2 could also cause reduction of cells survial associated with exposed doses and time and apoptosis.Conclusion Cadmium chloride coucld induce 293 cell apoptosis in vitro associated with exposed dose and time.
