流感大流行期病原学快速检测方法的建立

Establishment of Rapid Detection Method of Pathogen During Wide-epidemic Period of Influenza

  • 摘要: 目的为了确定1996年和1998年沈阳地区发生的严重流行性感冒的病原,建立快速、敏感的诊断流感的方法。方法用反转录-聚合酶链反应(RT-PCR)法,对门诊疑似流感病人咽拭子标本进行狗肾传代细胞(MDCK)短时培养后流感病毒基因检测。结果1996年流感流行期甲型流感病毒阳性率为60%;1998年流感流行时甲型流感病毒阳性率为83%,说明1996年和1998年流感大流行皆为甲型流感病毒所致。结论研究显示短时细胞培养后检测咽拭子标本中的流感病毒基因,为快速诊断流感的最佳途径。

     

    Abstract: ObjectiveTo deter min the pathogen of serious epidemic of influenza occurr ed in Shenyang area in 1996 and 1998,and for establishing rapid and sensitive method of diag nosis of influenza.MethodsThroat swabs were collected from the patients who were suspected to have influenza,and influenza virus gene was detected by reverse transcription-polymerase chain reaction(RT-PCT)after short culture in MDCK cell.ResultsPositive rate of influenza virus A was 60% in 1996 and 83% in 1998.Influenza virus A was considered to be the pathogenic agent of epidemic in 1996 and 1998.ConclusionRTPCRmethod after short cell culture was the best way for diagnosis of influenza.
